Background: People's knowledge and attitude play a role in deciding whether or not to use assisted reproductive procedures (ART). There is no information about people's attitudes and awareness about ART in western Iran. Thus, this study was performed to determine the knowledge and attitude of infertile people toward ART. Methods: This cross-sectional study was performed on 124 infertile participants who referred to the in vitro fertilization section of Fatemieh hospital, Hamadan, Iran. To measure the knowledge and attitude of the participants, 2 researcher-made questionnaires were used. Their validity and reliability were confirmed by content validity (eg, content validity index [CVI]; content validity ratio [CVR] ) and the Cronbach alpha, respectively. Results: The study involved 124 participants, more than half of whom were women (54.8%). The mean of the CVI and the CVR for the attitude questionnaire was 0.76 and 0.79, respectively, and for the knowledge, questionnaire was 0.72 and 0.71, respectively. The Cronbach alpha coefficient obtained for the attitude and knowledge questionnaires was 0.761 and 0.745, respectively. The mean (SD) of the attitude and knowledge of infertile people toward the use of ART was 15.33 ± 2.91 and 9.04 ± 2.92, respectively. The study reported that participants' age, inhabitation, and job status had a significant effect on their attitude score. Also, the study found that the knowledge score was inversely related to age. The knowledge in those who were unemployed and living in the rural and less than those employed and living in urban areas, respectively. Conclusion: The 2 questionnaires designed in this study (knowledge and attitude) have the required validity and reliability. As a result, it appears that increasing people's knowledge and improving their attitude, particularly among the elderly, is necessary.

OBJECTIVE: To investigate the effect of H2O2 on the migration and antioxidant defense of mesenchymal stem cells (MSCs) and the neurotrophic effects of H2O2-treated MSCs on spinal cord injury (SCI). RESULTS: Sublethal concentrations of H2O2 decreased cell migration and expression of CXCR4 and CCR2 as well as Nrf2 expression in MSCs. In the second phase, transplantation of treated and untreated MSCs to SCI caused minor changes in locomotor dysfunction. There was a significantly difference between cell-treated and spinal cord injury groups in expression of BDNF (brain-derived neurotrophic factor). Transplantation of H2O2-treated cells caused an increase in BDNF expression compared to non-treated cells. CONCLUSION: Transplantation of H2O2-treated stem cells may have protective effects against SCI through by increasing neurotrophic factors.

Recent evidence demonstrates that female subjects show exaggerated responses to 3,4-methylenedioxymethamphetamine (MDMA) compared with males. The aim of our study was to evaluate sex differences and the role of endogenous gonadal hormones on the effects of MDMA. Fifty-six intact and gonadectomized male and female Sprague-Dawley rats were randomly assigned to either MDMA (5 mg/kg) or saline treatment. Learning and memory were assessed using the Morris water maze (MWM). The expression of Bax and Bcl-2 in the hippocampus was detected by Western blotting. Behavioral analysis showed that MDMA led to memory impairment in both male and female rats. The female rats showed more sensitivity to impairment than the males, as assessed using all the memory parameters in the MWM. Ovariectomy attenuated the MDMA-induced memory impairment. By contrast, orchiectomized rats showed more impairment than MDMA-treated intact male rats. Bcl-2 and Bax were down-regulated and up-regulated in MDMA-treated male and female rats, respectively. MDMA treatment in the orchiectomized rats led to upregulation of Bax and down-regulation of Bcl-2. Ovariectomy attenuated the MDMA-induced up-regulation of Bax and caused more expression of Bcl-2 compared with what was observed in the MDMA-treated intact female rats. In summary, female rats showed exaggerated responses to the effects of MDMA and this may be explained by endogenous gonadal hormones.

We evaluated gene expression of estrogen and progesterone nuclear receptors in granulosa cells (GCs) of polycystic ovary syndrome (PCOS) women compared to women with normal cycling ovaries (control group) to achieve a better understanding of ovarian steroid status in patients with PCOS. In this prospective study, 40 patients with PCOS and 40 women with normal ovulatory function who underwent in vitro fertilization (IVF) for treatment of tubal and/or male infertility were recruited. Follicular fluid was collected from patients and GCs were isolated from follicular fluid and then were purified with Micro Beads conjugated to monoclonal anti-human CD45 antibodies. RNA was extracted and reverse transcription was performed. Gene expression of estrogen and progesterone receptors was determined by quantitative real time PCR (qRT-PCR). Estrogen receptor ß (ERß) expression was significantly higher than ERα expression in both groups (p < 0.002). ERα and ERß mRNA expression in PCOS was significantly lower than control group (p < 0.002). The expression levels of PRA and PRB in PCOS was significantly lower than control group (p < 0.002). In conclusion, a significant reduction of these genes in GCs from follicles of women with PCOS could be considered as a sign for maturation defect or follicular arrest in GCs.
